Ensimmäisen elokuvan lopussa AYT (Amerikan Yhdysvaltain taikakongressi) vangitsi voimakkaan pimeyden velhon Gellert Grindelwaldin (Johnny Depp) Lisko Scamanderin (Eddie Redmayne) avulla. Mutta toteuttaakseen uhkauksensa Grindelwald pakeni ja ryhtyi kokoamaan kannattajajoukkoa, josta valtaosa ei osaa aavistaakaan hänen todellisia aikeitaan: nostaa valtaan puhdasveriset velhot hallitsemaan kaikkia ei-taikovia. Estääkseen Grindelwaldin suunnitelmat Albus Dumbledore (Jude Law) värvää avukseen entisen oppilaansa Lisko Scamanderin, joka suostuu auttamaan tietämättömänä uhkaavista vaaroista.
